DFHO, a small-molecule analog of the intrinsic fluorophore of red fluorescent protein (RFP), is a cell permeable and non-cytotoxic fluorogenic high affinity ligand (Kd < 1 nM) of Corn aptamer that induces highly photostable yellow fluorescence (600-fold fluorescence). The S-adenosylmethionine (SAM) binding-induced dimerization of the SAM-Corn aptamer creates a binding pocket for its fluorogenic ligand DFHO, emitting a fluorescent signal that allows the monitoring of SAM levels in living cells.
