Asparaginase (ASNase, EC 3.5.1.1) is a type of amidase that catalyzes the hydrolysis of asparagine to aspartic acid and ammonia, playing an important regulatory role in nitrogen metabolism. Soil Asparaginase (S-ASNase) catalyzes the hydrolysis of asparagine in the soil to produce aspartic acid and ammonia. Under strong alkaline conditions, the ammonia reacts with hypochlorite and phenol to form a stable, water-soluble blue indophenol compound. This compound has a characteristic absorption peak at 630 nm, and the enzyme activity can be calculated by measuring the rate of increase in ammonia production.
