Introduction:Streptavidin is a tetrameric protein purified from Streptomyces avidinii that binds very tightly to the vitamin biotin with a dissociation constant on the order of ≈10⁻¹⁴ mol/L. The high affinity recognition of biotin and biotinylated molecules has made streptavidin one of the most important components in diagnostics and laboratory kits.The streptavidin fluorochrome conjugates are commonly used in indirect staining protocols to detect biotinylated primary antibodies in flow cytometry, microplate assays and microarrays.Conjugation:The APC was conjugated with Streptavidin under optimum conditions. Unconjugated streptavidin and mostly unconjugated APC were removed. The APC conjugates are prepared from chemically crosslinked APC to avoid dissociation of the molecule into subunits when highly diluted.Recommended Usage for Application:Flow: 0.02-1.25μg/mLApplication Note:Centrifuge protein conjugate solutions briefly in a microcentrifuge before use and use only the supernatant for the experiment. This step will eliminate any protein aggregates that may have formed in solution, thereby reducing nonspecific background staining.
