It's a multidomain molecule with an N-terminal PAN/APPLE-like domain, four Kringle domains, and a serine proteinase-like domain lacking detectable protease activity. Human HGF is secreted as an inactive single chain propeptide and cleaved to form a bioactive disulfide-linked molecule with alpha and beta chains. Alternate splicing generates human HGF isoforms with variations in the Kringle domains. Human HGF shares high amino acid sequence identity with other mammals. HGF binds to heparan-sulfate proteoglycans and the receptor tyrosine kinase, HGF R/c-MET. HGF-dependent c-MET activation is implicated in cancer development. HGF regulates epithelial morphogenesis by inducing cell scattering and branching tubulogenesis. It induces the up-regulation of integrin alpha 2 beta 1 in epithelial cells and can alter epithelium morphology. In the thyroid, HGF influences thyrocyte behavior and inhibits iodine uptake. HGF promotes the motility of cardiac stem cells in damaged myocardium.
