Pleuromutilin is a potent diterpene antibiotic with activity against gram-positive bacteria. The antibiotic was isolated in 1951 from the genus|Pleurotus.|It has been demonstrated through crystallization and biochemical research that Pleuromutilins acts by blocking substrate binding to the donor site and acceptor site of the ribosome's peptidyl-transferase center, which results in inhibition of the peptide bond formation. The compound encounters minimum cross-resistance with other compounds, which is attributed to its mode of action. Pleuromutilins targets a wide spectrum of pathogens and has been used in skin and respiratory tract infection studies.
