Nuclear Fast Red Staining Solution

Aladdin's Nuclear Fast Red Staining Solution can be used to stain cell nuclei with Nuclear Fast Red. It is commonly used as a counterstain for paraffin and frozen sections of different tissues. Nuclear Fast Red, also known as Kernechtrot or Calcium Red, has a molecular formula of C14H8NNaO7S with a molecular weight of 357.27 and a CAS No. of 6409-77-4. It is slightly red to dark brown powder soluble in ethanol and water. It is a counterstain commonly used to stain nucleus in red, and can also be used for the determination of calcium in serum (nuclear solid red colorimetric method). It is often used in conjunction with Prussian Blue Staining Solution.This product can be used directly without dilution and is reusable, but too many uses will result in less effective staining.This product is highly sensitive in staining samples, with low background production and clear staining results. Please refer to Figure 1 for the performance of this product in staining mouse testis paraffin sections.Figure 1. This figure is for reference only, which may vary due to different experimental conditions.When using 100µl of Staining Solution for each section, C0151-100ml is sufficient for staining 1000 sections.Precautions：Precipitation may occur in the Nucleus Fast Red Staining Solution at low temperatures. Under such circumstance, dissolve the precipitates completely at 25-50℃, and mix well before use.For the first time users of this product, it is recommended to perform a preliminary test with 1 or 2 samples to determine the optimal conditions for staining.It is recommended to stain the nucleus in a shallow way as long as the nucleus can be identified. Over-staining may impact the staining of other parts of cells.Nuclear fast red is soluble in water and ethanol, thus the washing time in water or dehydration time in ethanol shold be controlled properly. The mounting medium should not contain water and ethanol either for preservation of sections.This product is for R&D only. Not for drug, household, or other uses.For your safety and health, please wear a lab coat and disposable gloves during the operation.Instructions for Use：1.Sample preparation.a.For paraffin sections:For paraffin sections:Prepare paraffin sections using a conventional protocol that includes sample collection, fixation, washing, dehydration, clearing, wax immersion, embedding, slicing, and drying, etc.Dewax in xylene for 5-10 minutes;Replace with fresh xylene and dewax for another 5-10 minutes;Immerse in absolute ethanol for 5 minutes;Immerse in 90% ethanol for 2 minutes;Immerse in 80% ethanol for 2 minutes;Immerse in 70% ethanol for 2 minutes;Proceed to step 2 or step 3.b.For frozen sections:Prepare frozen sections using a conventional protocol that includes sample collection, freezing, slicing, and drying, etc.Wash with distilled water for 2 minutes;Proceed to step 2 or step 3.2.(Optional) Staining with other stains. 3.Nuclear Fast Red staining.a.Add 50-100µl of Nuclear Fast Red Staining Solution dropwise to each section, stain for 5-10 minutes, remove the staining solution, and wash section twice with tap water for 5 minutes each.Note: The staining time could be adjusted based on the actual staining result.b.Immerse sections in 70% ethanol for 10 seconds, 80% ethanol for 10 seconds, 90% ethanol for 10 seconds, anhydrous ethanol for 10 seconds, xylene for 5 minutes, and then mount slides with Neutral Resin. c.Observe and photograph under a microscope.
